I'm still on the fence regarding the game. What I've seen from the multiplayer (this post) looks pretty good, but the singleplayer is not what I'm getting it for, if I'm getting it.

That is an interesting development. Possibilities like this cross my mind from time to time ("They never said Ezio was descended from Altaïr.") but I usually dismiss them because of Occam's razor. The simplest explanation was that the bloodline from Altaïr to Desmond passed directly through Ezio. They're all Assassins, after all.

Not so fast! The notion that Altaïr is on Desmond's father's side of the family and Ezio's on his mother's side (or vice versa) is only one possibility. The first common descendant of Altaïr and Ezio could just as well be Desmond's father, or his grandfather, or his mother. It could even be Ezio's son if Ezio marries a great-great-granddaughter of Altaïr.
If Desmond is a descendant of both of them, then they're related through Desmond, technically...right?
You have two biological grandmothers, Joey. Assuming they're both alive and well... suppose they're having lunch together and someone asks how they are related. One of them might say, "This is Joey's other grandmother." or "My son married her daughter." There's no word for their relationship. You might say they're related "informally" or "socially" but a genealogist would say they're unrelated, technically.

When I heard what Revelations was about, I knew I heard about the handheld plans for Ezio travelling to Masyaf. I didn't say anything, because I thought I was mistaken. I doubt myself too much. haha

I'm with Stab on this, it does explain a lot. There is the issue about the Codex and how it got from Altair's to Marco Polo's possession. Altair mentions traveling to Mongolia to investigate Ghengis Khan, which is where Marco Polo found the Codex according to the writings in the Auditore crypt. I always assumed that Altair would've past the Codex down to is children so to keep it safe but maybe something happened.
I'm willing to bet that it's possible one of the Altair "seal" memories takes place in Mongolia and that could be where he died. That's just wild speculation though so i could be wrong.

Apparently Ubisoft worked closely with the author for this book but left the details up to him, then approved of them later. So the main events are most likely legitimate. Hopefully there will be differences in the game, or even exclusive details.

Again, so what? Most people don't, even today. Maybe it's me being used to genetic memory in this series, but I really feel Altair's twilight years don't need to be explored.

I expected the whole side-mission thing to not be totally gone, and it looks like templar agents are the main side-assassinations, which means we'll get lots of diverse-looking targets to prey upon, and that there will only be about 10 of them, so assassination mission designers will have to put more in the story.
I understood what they meant by saying that side-missions were gone from the start: that the ones that are really not worth even one single cut-scene or are relatively simple are now serving their true purpose (diversions for funz, like thif, assassination, and courtesan missions) and the more story-driven and/or cinematic/challenging ones are going to stay in (Templar Agents, Lairs, War Machine type missions).
I have no problem with that. Glad that I don't have to spend hours waiting in a menu if I just want to replay an short event where I beat up some schmo like in AC2.

The following interview with Darby McDevitt clarifies things a little. hewkii9's hunch was correct: Desmond has Assassin blood on both sides of his family. In fact, it looks like each of Desmond's four grandparents will descend from different Assassins.

Anyways, it's always good that a stealth game has disguises. Hopefully in a future game we'll be able to choose when we use one.
They should take a cue from Hitman and allow us to pick whatever outfit we want after knocking the person unconscious (don't want any blood on the new duds).
That's exactly what I was thinking after watching the Hitman Absolution gameplay.
Hopefully we'll get an Altair flashback where we dress up as a beggar, grab four homeless friends, and harass him for money until he kills himself.

Yeah, I didn't like that one mission in Bruddahood. Where you had to carry a chest full of money and dressed as a Captain.
Hopefully there will be less missions where it's timed and you feel like you're just running from point A to point B.
