The Surgeon Early Death
In the memory The Surgeon, you are first introduced to the target Silas in a scene. While playing through I discovered a way to assassinate Charles Lee. Position him so that he's around a corner. Then whistle for a guard. When the guard gets close enough on the other side of him, the weapon icon will light up to show you can do a corner assassination.
Be sure to use a smoke bomb so you won't get seen otherwise you'll fail the mission. I tried this on Charles earlier in the memory with no luck. Even if I had been successful, the game would have desynched me for an ally dying. I guess the game developers weren't worried about Charles dying in this part of the memory.
Blitz
The fastest way I could kill Silas. Right after the obligatory human shield tutorial, run straight to the fortress. There will be a brief lag when you get close from the game loading NPCs. climb up at the place shown and shoot Silas. Luckily I didn't have to move the camera since he's right in front of you, although you may target another enemy next to him or if enemies are passing close by (the chances of which I'm not sure of).
Dynamic Bullseye
Using the kill which involves impaling someone through with a bayonet and shooting another enemy in front of him. The target goes after you as soon as he is onscreen, so you need to act fast. It's dynamic because the target and Haytham are running. This is also useful in that you normally can't get non combat assassinations on enemies who have already detected you.
Easy Anonymous Kill

So escape any guards chasing after you by running into the haystack behind the fortress. After Silas comes after you, hide in the haystack again. The guards will walk back (sometimes Benjamin Church gets in a fight with them; let them settle their differences). The target won't detect you anymore.
Horse Assisted Assassination
Knocking someone down with a horse followed by a ground kill has been done before.

Again useful because of how the target detects you instantly.
Drowning
The challenge of moving Silas into water without damaging his health. I planned to use the water right by the fortress, but because of the target's AI and the fact that this was at the end of my recording, I settled for an easier place between a boat and a dock.

Did you know that we can throw coins during assassinate someone to avoid get desync in AC4? Does it possibility in AC3 as well?
You mean during a corner kill so the game doesn't know the character is dead? I'm sure it also works in AC3. Thing is, sometimes the game won't let you throw coins.
If you didn't whistle or use smoke bomb, can you assassinate Charles Lee?
As far as I know, a guard has to be within a certain distance for the assassinate option to appear. The smoke bomb was so I wouldn't get detected, or else mission failed.
Sorry it took so long to get this on the front page. We don't have many AC3 videos. This one shows much creativity.
