I like what Shaun says when you keep pestering him after the dialogue at the beginning. He will eventually say something like "Lucy, this new guy of yours, I think he's stalking me, he won't leave me alone."
Heck, half of what Shaun says makes me laugh.

I get annoyed every time I hear Shaun say that. Desmond should respond, "You mean like I'm going to be doing after this training while you continue to sit here on your ass?"

For a day or two while he trains. Then he'll be out there risking his life more than any other single person affiliated with the Assassins while Shaun stays home and continues to play desk jockey. Yet Shaun has the audacity to imply that Desmond's role is trivial while others are the real heroes. Talk about a double standard.
Shaun's statement was insulting and disrespectful, considering that Desmond was willing to make this huge sacrifice voluntarily. You don't ask someone to risk their life for your cause and then berate them five minutes later before they've even had a chance to begin. If it were me, I probably would have decked him for such a callous remark.
Well, Asaic, keep in mind that at this point in the training Desmond probably would get wrecked by Shaun. Even if he is a desk jockey, he's outright told Desmond he's killed before, and won't hesitate to do it again. Perhaps there will be an altercation in AC3 where Shaun and Desmond's tension finally boils over. Probably just fist fights, but then again, Desmond DOES have a hidden blade now (and a giant metal beating stick).

Having killed someone only speaks to his morals and his dedication to the cause. Even cowards and wimps can kill. He could have poisoned somebody's coffee, or snuck up behind them and shot them in the back of the head with a 9mm. Nothing about Shaun implies that he's trained in deadly martial arts. If he was, he would have been out there fighting the Abstergo goons instead of acting all panicky. He's a skinny nerd with an attitude and Desmond is pretty solid and athletic. I see no contest.
Besides, if Desmond decked him and Shaun decided to fight back with some mad kung fu, Lucy wouldn't let him. She'd sooner fire Shaun (or get involved herself) than let him "wreck" Desmond.
Anyways, I digress. I'm way off topic here.

For me, the music is more emotionally affecting than the dialogue. It's so melancholy.
But the dialogue bits that get to me the most are when Ezio is escorting his mom and sister out of Florence and when Claudia keeps questioning him, he has to keep saying "I don't know". Then he has to lie to them about the men being properly buried (I assume he's lying), getting a glimmer of how his life has to be from now on.
Finally, when he tells Rosa, "It's my birthday". That is just so sad.
Now I've gone and depressed everybody .

Yes, that is a truly epic line, I'm glad I finished the game before downloading the DLC, as that feels a lot like a conclusion to the game.

In terms of amusing quotes it has to be, "It's a me, Mario!" which has already been mentioned or perhaps when civilians shout "It's my lucky day!" in overly-Italian accents when you throw money.
I also love it when guards taunt you, saying something about how they'll soon be with your woman, before Ezio stabs them in a ruthlessly badass way with a counter-kill. Truly satisfying knowing he'll get nowhere near your woman
On a more serious note, much of Ezio's dialogue is pretty epic, especially many of the quotes in Latin/Italian (mainly during the Memory Corridors).
Oh and when citizens say stuff like "Life expectancy - I'd say about 3 months" as though they're an expert on life and death Sometimes the guards say, "What are you doing up here?! You belong on the ground, with the rats!" When they say that I climb back up and kill them.
From the first AC my favourites quotes would be:

I can't help but laugh when I remember that, since I haven't been able to recreate it. It's only happened once.
