This video shows a Beat Up even in San Gimignano. It shows that you can kill the woman who gives you the mission after you speak to her without a penalty, and how to do some weird poison trick I found ( but probably not first to discover). This works on all Beat Up events, after the cutscene they swagger around and you can kill them without penalty

Sort of related, in AC1 I always kill the pickpocket targets so they don't tell anyone they lost the package Crazy
