To perform this stunt you have to lure the target to a certain spot with a dead body. Then you can do a mini eagle strike (not as high as an eagle strike should be but high enough that the fall will kill you). Of course you could kill him anytime when he's out in the open but what could be cooler than jumping from the tallest spot in the abbey?

This is the walkthrough on setting up the eagle strike. Kill the 3 patrolling guards and make a trail of their dead bodies to lure Stefano to the eagle strike location. Enter the courtyard to trigger Stefano's movement and conversation ("Let us pray"). Climb up to the tower roof and hang from the very corner of the roof. I do this by walking toward the corner until Ezio starts to look down. Then I drop over the edge, and slide over so that both hands grab the corner.

Also included in this video is a stunt on Stefano. It is very similar to the eagle strike, but additional bodies are needed to lure Stefano to the correct location. The aim on the final jump is made by looking straight ahead, tilting the left stick directly to the left, and then jumping.
